Abstract
RNA helikaze su visoko konzervirani enzimi prisutni u sve tri domene života te u nekim virusima. Uključeni su u sve korake RNA metabolizma od transkripcije do degradacije. Uspješno razdvajaju kraće dlRNA, RNA-DNA heteroduplekse, remodeliraju strukturu RNA te uklanjaju RNA iz RNP kompleksa. Sve te funkcije obavljaju u ovisnosti o vezanju i hidrolizi ATP-a. U ovom radu detaljnije su opisane eukariotske RNA helikaze s DEAD domenom. Karakteristika svih je posjedovanje dvije RecA proteinu slične domene s RNA i ATP veznim mjestima u srži enzima. Na primjeru eIF4A i Dbp5 pokazano je kako ovi visoko konzervirani enzimi u suradnji s drugim proteinima mogu obavljati vrlo različite uloge kao što su inicijacija translacije te transport mRNA iz jezgre u citoplazmu.
Abstract (english)
RNA helicases are highly conserved enzymes abundant in all three life domains and some viruses. These enzymes participate in all stages of RNA metabolism from transcription to degradation and are very effective in unwinding short dsRNAs, RNA-DNA heteroduplexes, remodeling RNA structure and removing RNAs from RNP complexes. All these functions are done in an ATP-dependent fashion. In this work, eucaryotic DEAD box RNA helicases are described in detail. Characteristic of all DEAD box proteins are two RecA-like domains which harbour RNA and ATP binding sites. These highly conserved enzymes in cooperation with other proteins can perform different functions such as the initiation of translation (eIF4A) and mRNA transport from nucleus to cytoplasm (Dbp5).
