Abstract

Starenje je proces koji progresivno zahvaća sve organe i organske sustave. Jedan od najčešće korištenih modela u istraživanju procesa starenja je koža, a u ovom radu je korišten model štakora. Mnoga istraživanja pokazuju da je akumulacija starih stanica glavni uzrok promjena koje nastaju sa starenjem. Da bi testirali ovu hipotezu, potrebno je razviti metodu koja omogućava promjenu sastava stanica kože. Pri tome treba odgovoriti na mnoga pitanja kao što je sposobnost stanica da se nakon reimplantacije ugrade u tkivo štakora, da je broj i gustoća reimplantiranih stanica dostatan da izazove odgovarajući efekt te da njihova učestalost dozvoljava detekciju i analizu. Rezultati su pokazali da se reimplantirane stanice nakon izolacije i obilježavanja efikasno ugrađuju u kožu leđa štakora i to u značajnom postotku od gotovo 6% što u odgovarajućim eksperimentalnim uvjetima otvara mogućnost da se značajno utječe na fiziologiju i teksturu kože. Ova metoda također otvara mogućnost šire eksperimentalne i potencijalno terapeutske primjene

Abstract (english)

Aging is a process that progressively affects all organs and organ systems. One of widely used models in study of aging is skin and in this paper we used rat as a model. Studies have shown accumulation of senescent cells to be the main cause of changes that occur with aging. To test this hypothesis, it is necessary to develop a method for changing composition of skin cells. Many questions need to be answered, such as the ability of cells to incorporate into tissue after reimplantation, whether the number and density of reimplanted cells is sufficient to induce proper effect and whether their frequency allows detection and analysis. Results show that re-implanted cells after isolation and labeling effectively implant in dorsal skin of rats in a significant percentage of almost 6 %. This opens up the possibility of significantly affecting physiology and skin texture but also of wider experimental and potentially therapeutic applications.
