Abstract

Sustavno istraživanje polimorfije organskih spojeva, a posebno djelatnih tvari, u fokusu je znanstvenih istraživanja više od 50 godina. Izuzetno su dobro istraženi fizikalni parametri koji utječu i dovode do nastanka polimorfnih formi, te je uspostavljena metodologija njihove identifikacije i karakterizacije. S druge strane, malobrojna su sustavna istraživanja kojima se nastoji spoznati u kojoj mjeri određena onečišćenja imaju utjecaj na nastanak i varijabilnost polimorfnih formi. U nakani da se potvrdi hipoteza da određena onečišćenja ili međuprodukti koji nastaju prilikom priprave djelatnih tvari mogu imati utjecaj na varijabilnost polimorfije, kao modelni spoj odabran je entakapon. Priređena je biblioteka od 11 strukturno sličnih onečišćenja te većina u literaturi opisanih polimorfnih formi entakapona. Nakon toga je provedeno istraživanje u kojem je korištenjem metoda cijepljenja i obogaćenog uzorka ispitan utjecaj vrste i strukture onečišćenja na varijabilnost polimorfije. Ustanovljeno je da je taj utjecaj veći što su spojevi sličnije građe. Na temelju dobivenih podataka potvrđena je osnovna hipoteza ovog rada.

Abstract (english)

Systematic research of polymorphism of organic compounds and especially active substances, has been the focus of scientific research for more than 50 years. The physical parameters that lead to formation of certain polymorphic forms have been well researched and the methodology of identification and characterization has been established. On the other hand, there is very little systematic approach to determine the extent and impact of certain contaminants on the formation and variability of polymorphic forms. In order to confirm the hypothesis that certain impurities or intermediates used in the preparation may affect the variability of polymorphism, entacapone was selected as model compound. A library of 11 structurally related compounds and polymorphic forms of entacapone described in the literature were prepared. A study in which the influence of
impurities on the variability of polymorphism was examined using seed or spike methods. This effect was found to be greater with structuraly similar compounds. Based on collected data, the basic hypothesis of this thesis was confirmed.
