Abstract

Ravnoteža proteoma ključna je za život svih organizama. Glavnu ulogu u razgradnji proteina kod
eukariota ima sustav proteasoma 26S. Unatoč iznimnoj konzerviranosti kod eukariota, brojna
istraživanja ukazuju na složenost i dinamičnost ovog sustava u biljaka. Jedan od razloga je važnost
brzog prilagođavanja biljaka na promjenjivost okoliša. Proteasomalni kompleks u biljaka sastoji
se od središnje i regulatorne podjedinice koje funkcioniraju zajednički. U biljaka se proteini
usmjeravaju u razgradnju dodatkom ubikvitina. Kako bi se proteinu dodala ubikvitinska oznaka,
potrebni su enzimi ubikvitin ligaze E1, E2 i E3. Broj pojedinog tipa ligaza u stanici ovisi o funkciji
koju obavljaju, a najbrojnije su ligaze E3 koje pokazuju i najveću specifičnost prema supstratima
koje označavaju za razgradnju. Sustav proteasomalne razgradnje regulira različite procese u
biljnom razvoju, odgovoru na stres, signalizaciji i razvoju plastida. Ovaj rad pruža uvid u
kompleksnost sustava razgradnje proteina u biljaka te predstavlja nekoliko konkretnih primjera
uloge ovog sustava u biljnom metabolizmu.

Abstract (english)

The balance of the proteome is essential for the life of all organisms. The 26S proteasome system
is the main mechanism of protein degradation. Plant proteasomes play an irreplaceable role in
rapid adaptation of plants to environmental changes. The proteasomal complex in plants consists
of a central and a regulatory subunit that function together. In plants, proteins are directed to
degradation by the addition of ubiquitin. To add the ubiquitin tag to the protein, the ubiquitin ligase
enzymes E1, E2 and E3 are needed. The number of each type of ligase in the cell depends on the
function they perform, and the most numerous are the E3 ligases, which show the highest
specificity for the substrates they mark for degradation. The proteasomal degradation system
regulates various processes in plant development, stress response, signaling and plastid
development. This paper provides an insight into the complexity of the protein breakdown system
in plants and presents several concrete examples of the role of this system in plant metabolism.
