Abstract

Etiologija i patogeneza tumora testisa čovjeka ostaju do danas nerazjašnjene. Ograničenom
razumijevanju doprinosi i tendencija istraživanja primarno na humanom materijalu te in vitro
sustavima. Eksperimentalni teratokarcinom miša jedan je od priznatijih animalnih modela za
istraživanje tumora testisa. Međutim, opravdanost korištenja ovog modela temelji se
isključivo na morfološkom profiliranju. Cilj ovog istraživanja bio je po prvi puta na
molekularnoj razini sustavno analizirati i usporediti mišji model s humanim teratokarcinom,
te testirati opravdanost upotrebe ovog animalnog modela. U tu svrhu analizirani su biljezi
proliferacije, apoptoze, diferencijacije i matičnosti metodama imunohistokemije i Western
blottinga. Rezultati ovog istraživanja ukazuju na to da animalni model eksperimentalnog
teratokarcinoma miša vjerno modelira teratokarcinom čovjeka ne samo na morfološkoj već i
na molekularno biološkoj razini. Iz svega navedenog proizlazi da je eksperimentalni
teratokarcinom miša pogodan za potrebe istraživanja razvoja teratokarcinoma čovjeka na
molekularnoj razini, te da je njegova upotreba u znanstvenim istraživanjima opravdana.

Abstract (english)

The etiology and pathogenesis of human testicular tumors remain unclear to this day, in
part due to the trend of human material and in vitro systems research. Experimental mouse
teratocarcinoma is one of the recognized animal models for testicular tumor research.
However, the justification for using this model is based solely on the morphological
profile. The goal of this research was to systematically analyze and compare the mouse
model with human teratocarcinoma at the molecular level for the first time, and to test the
justification of its use in research. For this purpose, markers of proliferation, apoptosis,
differentiation and stemness were analyzed using immunohistochemistry and Western
blotting. The results indicate that the animal model of experimental mouse
teratocarcinoma faithfully models human teratocarcinoma not only at the morphological
but also at the molecular level. From all of the above, it follows that experimental mouse
teratocarcinoma is suitable for researching the development of human teratocarcinoma at
the molecular level, and that its use in scientific research is justified.
