Abstract

Geni glavnog sustava tkivne podudarnosti (MHC) imaju važnu ulogu u adaptivnoj imunosti, budući da kodiraju molekule koje limfocitima T prezentiraju antigene uzročnika bolesti. Najznačajnija karakteristika gena MHC je njihova izrazita varijabilnost, a smatra se da je glavna evolucijska sila koja pokreće i održava tu varijabilnost selekcija posredovana uzročnicima bolesti. Veliki američki metilj (Fascioloides magna) nezavičajni je nametnik u Europi čije nedavno širenje predstavlja prijetnju populacijama obične srne (Capreolus capreolus), jer kao aberantni nositelji u pravilu ne preživljavaju invaziju. U ovom istraživanju genotipiziran je lokus DRB skupine II gena MHC u 138 jedinki srne iz Hrvatske kako bi se istražila razina varijabilnosti. Ispitane su tri različite metode genotipizacije te je tehnologija sekvenciranja Illumina izabrana kao najoptimalnija. Broj utvrđenih alela (10), kao i indeksi genetičke raznolikosti ukazuju na umjerenu razinu varijabilnosti. Provedbom istraživanja prema principu „slučaj-kontrola“ uspoređeni su obrasci polimorfizma lokusa DRB s invazijom velikim američkim metiljem pri čemu je za alel Caca-DRB*0303 utvrđena statistički značajno (p < 0,01) veća prisutnost u skupini srna s područja na kojem je potvrđena fascioloidoza.

Abstract (english)

Major histocompatibility complex genes (MHC) have an important role in adaptive immunity since they code for molecules which present pathogen-dervied antigens to T lymphocytes. The most significant characteristic of MHC genes is their marked variability, and it is considered that the main evolutionary force that initiates and maintains this variability is pathogen mediated selection. The large American liver fluke (Fascioloides magna) is a non-native parasite in Europe whose recent spread poses a threat to the populations of roe deer (Capreolus capreolus), considering that roe deer as aberrant carriers usually do not survive the invasion. In this study, DRB locus of the MHC class II was genotyped in 138 roe deer from Croatia, in order to investigate the level of variability. Three different genotyping methods were tested and Illumina sequencing technology was chosen as the most optimal. The number of determined alleles (10), as well as the indices of genetic diversity, indicate a moderate level of variability. During our research conducted using the case-control method, polymorphism patterns at the DRB gene were compared with the occurrence of great American liver fluke infection, and it was found that the Caca-DRB*0303 allele exhibited a statistically significant (p < 0.01) higher presence in the group of roe deer from the area where fascioloidosis was confirmed.
