Abstract

Korištenjem poznatih podataka o dijelovima i procesima u stanici tokom diobe, postavili smo model dinamike između populacija stanica u raznim djelovima prometafaze, metafaze i anafaze. Rješavanjem pojednostavljenog modela uvidjeli smo važnost uvođenja vremenskog limita, kojeg uvodimo u obliku spindle assembly checkpoint-a (SAC-a). Rješavanjem općenitog modela uviđamo da populacije stanica sa više kromosoma sporije napreduju u formiranju diobenog vretena. Takve stanice kasne u metafazu i anafazu te imaju veći gubitak kromosoma. Ispitali smo neke od važnijih ili manje poznatih parametara kako bi ustanovili njihov utjecaj na trajanje formiranja diobenog vretena te gubitak kromosoma. Trajanje formiranja diobenog se povećalo sa povećanjem broja kromosoma i za male iznose parametra 𝛼, koji označava odnos broja mikrotubula i kromosoma, dok je geometrija stanica manje utjecajna. Nadalje smo ispitali parametre vezane za SAC te ustanovili važnost i utjecaj na gubitak kromosoma.

Abstract (english)

Using known information and data about cell parts and processes during cellular division, we made a model of population dynamics between cells in different stages of prometaphase, metaphase and anapahase. By solving the simplified model, we saw the importance of introducing a time limit, which we introduce in the form of the spindle assembly chepoint and the time where cells skip it. We solved the general model and found that populations of cells with more chromosomes form the spindle more slowly, and are thus late into metaphase and anaphase, and have a higher chomosome loss. We examined some of the more important or unknown parameters to find how they infulence the duration of the spindle assembly and chromosome loss. The duration of spindle assembly increases with the increase in the number of chromosomes in a cell, and with smaller values of the parameter 𝛼, which comes from the relation between the number of microtubules and chromosomes. The geometry affects the duration of spindle assembly less than the already listed parameters. Furthermore, we examined the parameters involved in modelling SAC, and determined their importance on chromosome loss.
