Abstract

Matične embrionalne stanice (ESC) su populacija stanica dobivena izolacijom iz unutarnje mase stanica (ICM) blastociste. One imaju sposobnosti pluripotencije i samoobnovljivosti. Ta dva svojstva čine ih popularnim predmetom istraživanja u razvojnoj biologiji, ali isto tako i nadom za moguću upotrebu u medicini. Zbog suprotstavljanja nekih zajednica ideji korištenja humanih embrija u znanosti, znanstvenici su uspjeli reprogramirati somatsku stanicu u pluripotentnu koristeći jedino transkripcijske faktore. Takve su stanice poznate pod nazivom inducirane pluripotentne matične stanice (iPSC) i gotovo su identične ES stanicama. Za bilo kakvu primjenu hESC ili hiPSC na ljudima, potrebno je razumjeti njihovu molekularnu osnovu. Brojna su se istraživanja, pogotovo u zadnjih nekoliko godina, bavila njihovim proučavanjem. Današnjim razvijenim tehnikama uzgoja i rukovanja ES, ali i iPS stanicama, sve se više produbljuje uvid u kompleksne mreže signalnih puteva i načine regulacije tih stanica i možda će, rješavanjem ograničenja jednih odnosno drugih vrsta stanica, one u bliskoj budućnosti postati dio široke primjene u medicini kao npr. u regenerativnoj terapiji i liječenju bolesti izazvanih načinom života.

Abstract (english)

Embryonic stem cells (ESC) are derived from the inner cell mass (ICM) of a blastocyst. These cells have properties like self-renewal and pluripotency which makes them a popular subject of research in developmental biology but also a hope for their use in, for example, regenerative therapies and the treatment of lifestyle diseases in the near future. In order for them to be applied in therapies, the molecular background has to be well understood. As some communities disapproved of using human embryos for scientific research, scientists succeeded in obtaining pluripotent cells from mature somatic cells using only transcriptional factors where the use of external pluripotent cells was not necessary. These cells are called induced pluripotent stem cells (iPSC). The understanding of ESC and iPSC are far from complete: advanced techniques in their cultivation and manipulation will provide a better insight into the complexity of signal networks and regulation systems of these cells and thus enable their wider application.
