Abstract

Posljednjih godina zapadna medicina sve se više okreće proizvodima iz prirode kao potencijalnim rješenjima za zdravstvene poteškoće koje se javljaju u današnjoj populaciji. Pčelinji otrov je jedan od tih proizvoda. Ima veliki potencijal za ublažavanje kroničnih bolova i koristi se za liječenje reumatskih bolesti, neuroloških bolesti i različitih dermatoloških stanja, te za liječenje tumora. Cilj ovog istraživanja je bio procijeniti učinak visoke koncentracije pčelinjeg otrova (100 μg/ml) na ljudske limfocite periferne krvi u uvjetima in vitro tijekom različitih vremenskih perioda (od 10 min do 24 h). Parametri komet-testa ukazuju na genotoksični učinak pčelinjeg otrova na ljudske limfocite periferne krvi u odnosu na kontrolni, netretirani uzorak i to u vremenskom izlaganju od 30 minuta, 6 i 24 sata za dužinu repa, te od 6 i 24 sata za repni moment i intenzitet repa. Rezultati mikronukleus-testa također ukazuju na genotoksični učinak visoke koncentracije pčelinjeg otrova na ljudske limfocite periferne krvi u odnosu na kontrolni, netretirani uzorak. Rezultati učestalosti pojavljivanja mikronukleusa, pupova i nukleoplazmatskih mostova pokazali su statistički značajno povećanje (p<0,05) u svim vremenima tretiranja u odnosu na kontrolni, netretirani uzorak, dakle utvrđena je ovisnost o vremenu. Rezultati dobiveni ovim radom upućuju na to da visoka koncentracija pčelinjeg otrova uzrokuje staničnu nestabilnost. Buduća istraživanja bilo bi korisno usmjeriti na dublje razumijevanje mehanizma djelovanja pčelinjeg otrova i njegovih sastavnica na ljudske stanice u svrhu primjene ovog prirodnog proizvoda u medicini.

Abstract (english)

Western medicine is turning more and more towards natural products as potential solutions for the health problems that occur in human's population. Bee venom is one of those products. It has great potential for chronic pain relief and it is used for treatment of rheumatic diseases, neurological diseases and various dermatological conditions and also for treatment of tumors. The aim of this study was to assess the effect of high concentration of bee venom (100 μg/ml) on human peripheral blood in vitro over a range of time spans (from 10 min to 24 h). Parameters of comet assay showed genotoxic effect of bee venom on human peripheral blood lymphocytes in comparison with control in time period of 30 min, 6 and 24 h for tail length and of 6 and 24 h for tail moment and tail intensity. Results of micronucleus test also showed genotoxic effect of high concentration of bee venom on human peripheral blood lymphocytes in comparison with control. Method parameters (number of micronucleus, nuclear buds and nucleoplasmic bridges) showed statistically significant increase (p<0,05) for all lengths of exposure in comparison with control, therefore time dependence was established. Results obtained by this study indicate that high concentration of bee venom can lead to cellular instability. Future research should be focused on deeper understanding of the mechanism of action of bee venom and its components on human cells and finding the best possible application of this natural product in medicine.
