Abstract

Embrionalne matične stanice se dobivaju iz unutarnje mase stanica blastociste, preimplantacijskog stanja embrija. One su pluripotentne, što znači da mogu diferencirati u više od 200 tipova stanica ljudskog tijela. Također, pod određenim uvjetima, embrionalne matične stanice se mogu neograničeno dijeliti. Zbog svoje plastičnosti i neograničene sposobnosti samoobnavljanja, embrionalne matične stanice se mogu koristiti u regenerativnoj medicini i tkivnom inženjerstvu nakon ozljede tkiva ili bolesti. Znanstvenici su uspješno dobili oligodendrocitne progenitorske stanice iz ljudskih embrionalnih matičnih stanica koje bi se mogle koristiti u liječenju ozljeda leđne moždine, dopaminergične neurone za Parkinsonovu bolest, astrocite i motoričke neurone za amiotrofičnu lateralnu bolest i pigmentni epitel za pacijente sa Stargardtovom makularnom distrofijom. Ovi primjeri pokazuju koliko su istraživanja na embrionalnim matičnim stanicama bitna. Uz etička pitanja o terapijama matičnim stanicama, postoje i neki tehnički problemi kao što su imunološka nekompatibilnost i formiranje teratoma. Dobivanjem embrionalnih matičnih stanica terapijskim kloniranjem mogu se riješiti etička pitanja i problem histokompatibilnosti. Ostale potencijalne koristi embrionalnih matičnih stanica podrazumijevaju istraživanja ranog ljudskog razvoja, proučavanje genetički nasljednih bolesti i „in vitro“ toksikološka istraživanja.

Abstract (english)

Embryonic stem cells are derived from the inner cell mass of a blastocyst, an earlystage preimplantation embryo. They are pluripotent, which means that they are able to differentiate into more than 200 cell types in the adult body. Additionally, under defined conditions, embryonic stem cells are capable of propagating themselves indefinitely. Because of ther plasticity and unlimited capacity for self-renewal, embryonic stem cell therapies have been proposed for regenerative medicine and tissue replacement after injury or disease. Scientist successfully derived oligodendrocyte progenitors from human embryonic stem cells that could be used in treating spinal cord injuries, dopamin producing neurons for Parkinson's disease, astrocytes and motor neurons for amiotrophic lateral disease and retinal pigment epithelium for patients with Stargard't macular distrophy. These examples show how important research on embryonic stem cells really are. Besides the ethical concerns of stem cell therapy, there are some techical problems of graft-versus-host disease and forming teratomas. However, ethical problems and graft-versus-host disease can be solved using therapeutic cloning. Other potential uses of embryonic stem cells include investigation of early human development, study of genetic disease and as „in vitro“ systems for toxicology testing.
