Abstract

Oksidacijski stres je u današnje vrijeme vrlo popularna tema budući da smo okruženi raznim okolišnim zagađivačima, kemikalijama i stresnom načinu života. Također je sve važniji zbog velike korelacije sa razvojem bolesti i ubrzanim starenjem. Osim u medicini važan je u okolišnim istraživanjima za ispitivanje kemikalija i ksenobiotika koristeći testne organizme i u genetičkom inženjerstvu za proizvodnju transgeničnih organizama otpornih na stres. Zbog toga je potrebno moći precizno izmjeriti razinu oksidacijskog stresa u organizmu. Dok su direktne metode koje mjere koncentraciju slobodnih radikala ograničene samo na uvjete in vitro postoje brojne indirektne metode koje mjere aktivnost Najvažniji biomarkeri oksidacijskog stresa su količina MDA te aktivnosti enzima SOD i GPX. Međutim, za potpunu sliku je najbolje koristiti nekoliko različitih parametara koji daju uvid u potpuni redoks status organizma jer antioksidacijski sustavi djeluju simultano i sinergistički.

Abstract (english)

Today oxidative stress is a very popular subject in science due to pollutants in our environment and stressful way of life. Moreover, it is very important because of great correlation with ageing and disease development. Apart from medicine, oxidative stress is measured in environmental studies using test organisms as indicators of xenobiotic toxicity and in genetic engineering for producing transgenic organisms with improved tolerance to stress. Because of its many uses, it is necessary to precisely measure level of oxidative stress in organisms. While direct methods, which measure concentration of free radicals, function only in in vitro conditions, there are plenty of indirect methods which measure antioxidant enzyme activities or by-products of damage by ROS on cellular components. The most reliable biomarkers of oxidative stress are MDA content and SOD and GPX activities. However, for complete insight in redox status it’s best to measure many parameters because antioxidant systems work simultaneously and synergistically.
