Abstract

Serotonin je biološki aktivan indolamin prisutan u mozgu i perifernim tkivima sisavaca. U ovom radu laboratorijski štakori su tretirani prekursorom sinteze serotonina, 5-hidroksitriptofanom (5HTP) ili neselektivnim inhibitorom monoamin-oksidaze, tranilciprominom (TCP), kako bi se uzrokovalo povišenje razine serotonina i promjena njegove homeostaze i zatim ispitao utjecaj povišene razine serotonina na jetru i bubrege. Eksperimentalnom tretmanu su podvrgnute trudne ženke te mladunci iz okota koji su žrtvovani kako bi se analizirali njihovi organi. Analizirani su histološki preparati jetre (N =34) i bubrega (N =38) te su morfometrijski izmjerene ciljne strukture u tim organima, koje su uključivale mjerenja promjera jezgri stanica, centralnih vena i sinusoida hepatocita unutar jetre te mjerenje površine bubrežnih tjelešaca, glomerula i proksimalnih kanalića bubrega. Rezultati su pokazali značajne statističke razlike između kontrolne skupine i životinja s povišenom razinom serotonina, pri čemu su životinje tretirane s TCP-om pokazale veća odstupanja u građi jetre, dok je 5HTP imao veći utjecaj na bubrege.

Abstract (english)

Serotonin (5HT) is a biological active indolamine in brain and peripheral tissues of the mammals. In this experiment rats were treated with serotonin synthesis precursor, 5 hydroxytryptophan (5HTP) or with nonselective monoamine oxidase inhibitor, tranylcypromine (TCP) in order to increase the level of serotonin and alter its homeostasis, and to investigate the influence of increased serotonin on the liver and kidney. The experiment started with treatment of pregnant females, continued on offsprings, and finished with immolation of baby rats, and isolaton of their organs for analysis. We have analysed tissue sections of liver (N =34) and kidneys (N =38), and morphometrically measured target structures in these organs, which included diameter of nucleus, cells, central venes and hepatic sinusoids and surfaces of the renal bodies, glomerules and proximal tubules in the kidney. Our results showed statistically significant differences between control group and rats with altered serotonin homeostasis. Large structural changes were recognised in the liver of animals treated with TCP, while rats treated with 5HTP displayed more severe structural changes in kidneys.
