Abstract

Pogreške u podjeli kromosoma za vrijeme stanične diobe uzrokuju aneuploidiju, stanje neravnoteže u broju kromosoma. Takvo stanje dovodi do teških bolesti i ubrzava stvaranje tumora. Stoga su otkrivanje podrijetla pogrešaka i njihov dugoročni učinak na stanice ključni za razumijevanje aneuploidije. Da bi se razumjeli mehanizmi koji dovode do velike učestalosti pogrešaka, uveli smo teorijski opis diobe stanica. U pojedinoj diobi, model opisuje mogućnost gubitka kromosoma i brzinu staničnog ciklusa, pri čemu je uzeto u obzir da obje pojave ovise o broju kromosoma u stanici. U modelu se prati veliki broj staničnih dioba gdje se svaka od tih dioba pojedinačno simulira. Na taj način daje se veza između procesa koji se događaju za vrijeme jedne diobe te izgleda cjelokupne populacije stanica. Također, predviđa se broj kromosoma u stanicama kroz generacije i vrste stanica koje prevladavaju nakon velikog broja dioba.

Abstract (english)

Dividing cells that experience chromosome missegregation generate aneuploid daughter cells, which contain an incorrect number of chromosomes. This causes severe diseases and promotes tumorigenesis. Hence, the discovery of causes and cellular consequences of missegregation is the key in understanding aneuploidy. To provide insight into important cell mechanisms that drive chromosome instability, we developed theoretical model of cell division. For every single cell division, the model describes the ability of chromosome gain/loss and cell proliferation, where it is taken into consideration that both of these processes depend on the cell karyotype. Model tracks a large number of cell divisions where every single division is simulated separately. In that way, the correlation between the processes in one single cell and the appearance of the whole population is achieved and studied. Likewise, the expected number of chromosomes in cells is predicted.
