Abstract

Karcinom debelog cijeva ubraja se među najčešće karcinome u oba spola te ima veliku stopu smrtnosti. Nastaje iz prethodno postojećih benignih tumora, odnosno adenoma. Uklanjanjem adenoma, bolest je lako prevenirati, a liječenje je moguće ukoliko je do dijagnoze došlo prije posljednjeg stadija, metastazirajućeg karcinoma.
U ovom seminaru izložena su dva puta karcinogeneze debelog crijeva, put gubitka heterozigotnosti i put mikrosatelitne nestabilnosti. Oba puta karakterizirana su gubitkom funkcionalnosti gena za tumorske supresore i aktivacijom onkogena te su poremećeni isti signalni putovi u stanici. Mutacije koje dovode do tih događaja razlikuju se ovisno o tipu karcinogeneze.

Abstract (english)

Colorectal cancer is one of the most common cancers in both sexes and has high mortality rate. It arises from previously existing benign tumors, adenomas. By removing the adenomas, the disease could be prevented, and treatment is possible if carcinoma is diagnosed before the last stage, metastatic cancer.
Two colon carcinogenesis pathways are showed in this seminar, a pathway based on loss of heterozygosity and a pathway based on microsatellite instability. Both pathways are characterized by loss of tumor suppressor gene function and oncogene activation, and both disrupt the same signaling pathways in the cell. The mutations that lead to these events vary depending on the type of carcinogenesis.
