Abstract

Shizofrenija je psihički poremećaj kojeg karakteriziraju 3 vrste simptoma: pozitivni, negativni te kognitivni. Patofiziološki mehanizam još nije u potpunosti razjašnjen, a istraživanja se grupiraju u nekoliko različitih hipoteza i područja istraživanja. Neke od tih hipoteza su dopaminergička hipoteza, koja se temelji na poremećenoj dopaminergičkoj neurotransmisiji, glutamatergička hipoteza, koja se temelji na hipofunkcionalnosti NMDA receptora, serotoninska hipoteza, GABAergična hipoteza itd. Još dva područja istraživanja jesu utjecaj upale i oksidativnog stresa na razvoj shizofrenije te utjecaj poremećene epigenetske regulacije u moduliranju ispoljavanja nasljeđenih rizičnih genetskih varijanti. U ovom seminaru opisana su dosadašnja istraživanja i njihovi rezultati u sklopu svake od navedenih hipoteza i područja istraživanja. Dosadašnji rezultati vrlo su raznoliki i potrebna je integracija podataka dobivenih istraživanjem pojedinih hipoteza u jednu cjelinu.

Abstract (english)

Schizophrenia is a mental disorder which is characterized by 3 types of symptoms: positive, negative and cognitive. Its pathophysiologic mechanism is not fully understood yet, but current research can be grouped into few different hypotheses and fields of research. Some these hypotheses are dopaminergic hypothesis, which is based on disrupted dopaminergic neurotransmission, glutamatergic hypothesis, which is based on hypofunctionality of NMDA receptors, serotoninergic hypothesis, GABAergic hypothesis, etc. Two more fields of research are influence of inflammation and oxidative stress on the development of schizophrenia and influence of disrupted epigenetic regulation in modulation of the effect of inherited genetic risk variants. In this work research so far and its results of these hypotheses and fields of research are described. So far results are very heterogeneous and there is need for integration of these results in a one big complex.
