Abstract

Humani leukocitni antigeni (HLA) reguliraju imunološke reakcije. Dosadašnja istraživanja utvrdila su povezanost polimorfizama gena sustava HLA s nastankom autoimunih bolesti. Antieritrocitna autoprotutijela (AP) mogu uzrokovati autoimunu hemolitičku anemiju (AIHA) koja se češće javlja u osoba s limfoproliferativnim i autoimunim bolestima i uzrokuju smanjenje životnog vijeka eritrocita. Cilj ovog doktorskog rada bio je ispitati moguću povezanost različitih alela sustava HLA razreda I i II s nastankom antieritrocitnih autoprotutijela razreda IgG u bolesnika i trudnica. Usporedbom rezultata HLA PCR-SSP (engl. Polymerase Chain Reaction - Sequence Specific Primer) tipizacije uzoraka bolesnika i trudnica s AP-ima razreda IgG, te zdravih ispitanika bez AP, utvrđena je moguća povezanost alela B*57 i DQB1*03:03 s pojavom antieritrocitnih autoprotutijela razreda IgG. Ovim istraživanjem nije otkriven protektivni alel.

Abstract (english)

Human leucocyte antigens (HLA) regulate an immune response. Previous studies have established a correlation of HLA polymorphisms with the development of autoimmune diseases. Antierytrocite autoantibodies (AB) may result in occurence of autoimmune haemolytic anaemia (AIHA), which is more frequent in individuals with lymphoproliferative and autoimmune diseases, and cause a decreased lifetime of erythrocytes. The aim of this doctoral thesis was to investigate the possible association of different HLA class I and II alleles with the development of RBC (engl. Red Blood Cell) autoantibodies of IgG class. Comparing the results of HLA PCR-SSP (engl. Polymerase Chain Reaction - Sequence Specific Primer) typing of patients with AB of IgG class and healthy individuals without AB, we found that alleles B*57 and DQB1*03:03 are susceptible for RBC AB development. In this investigation we did not observed any protective allele.
