Abstract

Karcinom debelog crijeva nastaje kao rezultat stupnjevitog procesa akumulacije genetičkih i epigenetičkih promjena u nizu onkogena i tumor supresorskih gena. Angiogeneza je proces neophodan za nastanak i napredovanje tumora, a faktor rasta vaskularnog endotela (VEGF) je jedna od ključnih molekula u ovom procesu. Budući da na razinu ekspresije gena VEGF utječu i regulatorni SNP polimorfizmi, cilj ovog istraživanja bio je ispitati ulogu ovih polimorfizama u sklonosti obolijevanju i preživljenju od sporadičnog karcinoma debelog crijeva. U analizi polimorfizama VEGF -634 G/C i VEGF +936 C/T korištena je metoda TaqManTM real-time PCR. Nije opažena statistički značajna razlika u raspodjeli pojedinih genotipova i haplotipova u skupini oboljelih od sporadičnog karcinoma debelog crijeva u odnosu na zdravu populaciju. Analiza preživljenja je pokazala da su oboljeli kod kojih je utvrđena prisutnost alela C polimorfizma -634 C/G živjeli duže (p=0.01), dok kod polimorfizma +936 C/T gena VEGF nije bilo statistički značajne razlike.

Abstract (english)

Colon cancer arises from a stepwise accumulation of genetic and epigenetic changes in the number of oncogenes and tumor suppressor genes. Angiogenesis is necessary for the formation and progression of tumors, and vascular endothelial growth factor (VEGF) is one of its key mediators. Since the VEGF expression can be affected by regulatory SNP polymorphisms, objective of this study was to investigate the role of these polymorphisms in the susceptibility to and survival of patients with sporadic colorectal cancer. VEGF polymorphisms -634G/C and +936 C/T were genotyped using TaqManTM real-time PCR. No association was found between genotypes and haplotypes and susceptibility to sporadic colorectal cancer. Survival analysis showed that patients with established presence of VEGF -634 C allele lived longer (p=0.01), while the +936 C/T VEGF polymorphism had no influence on patients’ survival.
