Abstract
Metoda CRISPR-Cas9 značajno je unaprijedila pristup uređivanju genoma. Iako je njezina primjena na genom jezgre danas iscrpno istražena, primjena na mitohondrijski genom i dalje predstavlja izazov. Glavni problem je unos strane, sgRNA molekule u matriks mitohondrija kroz dvije mitohondrijske membrane. Iako su mehanizmi prijenosa sgRNA i dalje nepoznati, u ovom radu predstavljeni su pristupi koji su rezultirali relativno uspješnim uređivanjem genoma mitohondrija. Daljnje unaprjeđenje i usavršavanje metoda za uređivanje mtDNA može pridonijeti potencijalnom liječenju mitohondrijskih bolesti te sprečavanju njihova prijenosa na potomstvo.
Abstract (english)
CRISPR-Cas9 method brought a significant progress to the genome editing. Although its application to the nuclear genome has been extensively studied, its application to the mitochondrial genome remains a challenge. The main problem is the entry of foreign, sgRNA molecules into the mitochondrial matrix across two mitochondrial membranes. Although the mechanisms of sgRNA transfer are still unknown, this manucript presents approaches that have resulted in relatively successful mitochondrial genome editing. Further improvement of the application of CRISPR-Cas9 on mtDNA can greatly contribute to the treatment of mitochondrial diseases and prevent their transmission to offspring .
