Abstract

Podzemlje dinarskog krša stanište je mnogih endemičnih i reliktnih svojti te predstavlja vruću
točku bioraznolikosti na globalnoj razini. Međutim, mikrobne zajednice dinarskog krškog
podzemlja slabo su istražene. U ovom radu su po prvi put opisane mikrobne zajednice endemskih
sesilnih filtratora: sve tri vrste dinarskog špiljskog školjkaša (Congeria jalzici, C. kusceri i C.
mulaomerovici) i dinarskog špiljskog cjevaša (Marifugia cavatica). Uzorkovana su tkiva školjkaša,
obraštaj njihove ljušture, tkiva cjevaša i voda s četiri lokacije u Hrvatskoj i Bosni i Hercegovini.
Bakterijski taksoni prisutni u uzorcima detektirani su sekvenciranjem fragmenata 16S rRNA gena
Illumina MiSeq metodom te bioinformatičkim analizama QIIME2 platformom. Ukupno je
detektirano 43 bakterijska koljena s koljenom Proteobacteria kao najzastupljenijim i 373 porodice.
Nadalje, utvrđene su značajne razlike u sastavu i abundanciji mikrobnih zajednica različitih tipova
uzoraka: tkiva i obrisaka školjkaša, tkiva cjevaša i vode. Mikrobne zajednice tkiva cjevaša značajno
su se razlikovale ovisno o lokaciji uzrokovanja, a mikrobne zajednice tkiva školjkaša nisu se
značajno razlikovale ni ovisno o vrsti ni ovisno o lokaciji uzorkovanja. Zaključno, ovaj rad donosi
prve podatke o mikrobnim zajednicama sesilnih filtratora dinarskog krškog podzemlja i kao takav
predstavlja temelj za daljnja istraživanja hranidbenih mreža te odnosa mikrobnih zajednica i
njihovih domaćina u dinarskom krškom podzemlju.

Abstract (english)

The Dinaric karst underground is a global biodiversity hotspot inhabited by many endemic and
relict taxa. However, microbial communities of Dinaric karst underground are significantly
understudied. This thesis for the first time describes the microbial communities of all three Dinaric
cave mussel species (Congeria jalzici, C. kusceri and C. mulaomerovici) and Dinaric cave
tubeworm (Marifugia cavatica). Tissues of mussels and tubeworms, biofilm of mussel shells, and
water were sampled at four locations in Croatia and Bosnia and Herzegovina. Bacterial taxa were
identified by 16S rRNA gene sequencing and QIIME2 bioinformatic analyses. A total of 43
bacterial phyla, with Proteobacteria as the most prevalent, and 373 families were detected.
Significant differences in the composition of microbial communities between different sample
types were found, i.e., tissue of mussels and tubeworms, biofilm of mussel shells, and water.
Tubeworms’ microbial communities differed significantly depending on the location, while the
microbial communities of mussels were not significantly different depending on the species or the
location. In conclusion, this thesis presents the first data on the microbial communities of sessile
filtrators in the Dinaric karst underground and is therefore a basis for further research of food webs
and host-microbe interactions in the Dinaric karst underground.
