Abstract

Virus pjegavosti i venuća rajčice (Tomato spotted wilt virus, TSWV) identificiran je prvi put 1930.
godine u Australiji kao uzročnik bolesti pjegavosti i venuća rajčice (Solanum lycopersicum L.).
Tijekom 2021. i 2022. godine su u Hrvatskoj zabilježeni simptomi TSWV-a na kultivarima rajčica
koji se reklamiraju kao rezistentni od strane proizvođača sjemena. Cilj ovog rada bio je potvrditi
TSWV kao uzročnika infekcije različitih biljaka s terena i odrediti biološke i molekularne
karakteristike virusnih izolata. Infekcija TSWV-om potvrđena je kod tri uzorka metodom reverzne
transkripcije i lančane reakcije polimerazom (RT-PCR). Filogenetskim analizama amplikona
pokazano je da se radi o izolatima virusa koji potječu iz regionalne virusne populacije. Analizom
aminokiselinskog slijeda za nestrukturni virusni protein NSm kod jednog izolata pokazano je da se
radi o varijanti virusa koja ne sadrži mutacije povezane s probijanjem rezistencije kod rajčice.
Biološki test kojim se nastojalo istražiti biološke značajke (krug domaćina, simptomatologija i
probijanje rezistencije kod rajčice) izolata TSWV-a nije bio uspješan zbog termolabilnosti virusa i
gubitka infektivnosti biljnog materijala uslijed neadekvatnih uvjeta pohrane.

Abstract (english)

Tomato spotted wilt virus (TSWV) was first identified in 1930. in Australia as the causative agent
of the spotted wilt disease of tomato (Solanum lycopersicum L.). In Croatia, during 2021. and
2022. symptoms of TSWV were observed on tomato cultivars marketed as resistant by seed
suppliers. The aim of this study was to confirm TSWV as the agent infecting various field plants
and to determine the biological and molecular characteristics of virus isolates. Infection with
TSWV was confirmed in three isolates by reverse transcription and polymerase chain reaction
(RT-PCR). Phylogenetic analyses of amplicons have shon that the virus isolates originated from
the regional viral population. The nonstructural protein NSm amino acid sequence analysis from
one isolate revealed the absence of mutations linked to the resistance-breaking phenotype in
tomato. The bioassays conducted to examine biological characteristics (host range,
symptomatology, resistance-breaking phenotype in tomato) of TSWV isolates was unsuccessful
due to thermal inactivation of the virus and the loss of infectivity in inadequately stored plant
material.
