Abstract

Virus uvijenosti lista vinove loze 3 (grapevine leafroll-associated virus 3, GLRaV-3,
Closteroviridae) uzročnik je bolesti uvijenosti lista, jedne od ekonomski najznačajnijih viroza
vinove loze. U ovom istraživanju, u uzorcima 16 autohtonih dalmatinskih sorti i klonskih
kandidata sorte 'Plavac Mali', primjenom molekularnih metoda napravljen je pregled
pojavnosti ekonomski štetnih virusa vinove loze uz poseban osvrt na genomske varijante
GLRaV-3. Utvrđena je dominantna pojavnost GLRaV-3 te učestale koinfekcije s drugim
virusima. Analizom populacijske strukture virusa GLRaV-3 utvrđena je pripadnost virusnih
varijanti filogenijskim skupinama I i II. Nakon inokulacije standardnih indikatorskih biljaka i
autohtone sorte 'Tribidrag' virusom GLRaV-3 samostalno ili u koinfekciji utvrđene su promjene
u relativnoj koncentraciji GLRaV-3 ovisno o starosti biljke, fenofazi dormancije i prisutnosti
drugih virusa. Pojava i jakost simptoma, kao i fiziološke promjene biljaka vinove loze, ovisile
su o sorti i sastavu virusnog inokuluma, pri čemu su se simptomi uvijenosti lista razvili na svim
sortama vinove loze.

Abstract (english)

Grapevine leafroll-associated virus 3 (GLRaV-3, Closteroviridae) is the causal agent of
grapevine leafroll disease, one of the most important diseases affecting grapevine. The
prevalence of economically harmful grapevine viruses was determined, with special reference
to genomic variants of GLRaV-3. In the sample of 16 indigenous Dalmatian varieties and clonal
candidates of 'Plavac Mali', the predominance of GLRaV-3 and its frequent coinfections were
determined. Population structure analysis of GLRaV-3 variants confirmed their clustering into
phylogenetic groups I and II. To better elucidate GLRaV-3 and its effect on the host, after
inoculation ofstandard indicator plants and indigenous variety 'Tribidrag' with GLRaV-3 singly
or in coinfection, differences in the relative GLRaV-3 concentration were determined as a
result of plant-host age, dormancy stage and interactions with other viruses. The appearance
and severity of symptoms as well as physiological changes were variety and inoculum
dependant, while leafroll symptoms developed in all grapevine varieties.
