Abstract

Bakteriofagi su virusi koji inficiraju bakterije i moguće ih je koristiti u borbi protiv bakterija
rezistentnih na antibiotike. Terapija fagima ne treba nužno zamijeniti antibiotike u liječenju već
se može koristit zajedno s njima čime se postiže sinergistički efekt. Kako bi fage dostavili
sigurno na mjesto infekcije koriste se različiti tipovi pripravaka – tekući, suhi (u obliku praška)
i pripravci s inkapsuliranim fagima. Svi oblici pripravaka imaju svoje prednosti i nedostatke te
se daljnja istraživanja trebaju usredotočiti na poboljšanje stabilnosti, učinkovitosti i sigurnosti
isporuke. Zasada, inkapsulacija faga u liposome, hidrogel ili na površinu vlakana pokazuje
obećavajuće rezultate . Omogućava fagima dolazak na mjesto infekcije, lakšu penetraciju u
tkiva i štiti ih od razgradnje. Trebalo bi dodatno istražiti i razmotriti mogućnost razvoja
sintetskih faga koji bi imali promjenjivu domenu za vezanje na receptore. Takve fage bi mogli
koristiti u terapiji protiv širokog spektra bakterijskih infekcija.

Abstract (english)

Bacteriophages are viruses that infect bacteria. Bacteriophages can be used to treat diseases
caused by antibiotic-resistant bacteria. That does not mean that phage therapy should replace
antibiotics instead, it can be used with them which results in a synergetic effect. To ensure
phages are safely delivered to the site of infection, various types of preparations are used –
liquid, solid (in powder form) and preparations with encapsulated phages. Each form of
preparation has its advantages and disadvantages and further research should focus on
improving stability, effectiveness and delivery safety. Currently, the encapsulation of phages
in liposomes, hydrogels and on the fiber surface shows the most promising results. These
types of preparations allow phages to reach the site of infection, protects them from
degradation and facilitates tissue penetration. Additional research and consideration should be
given to the development of synthetic phages with variable receptor-binding domains. Such
phages could be used in therapy against a broad spectrum of bacterial infections.
