Abstract

Prisutnost zajednica fitoplanktona i rasprostranjenost fotosintetskih pigmenata utvrđivani su u dva
prirodna krška jezera, u Vranskom jezeru kod Biograda i Vranskom jezeru na Cresu. Cilj ovog rada
bio je usporediti metodu tekućinske kromatografije visoke djelotvornosti (HPLC) s taksonomskom
identifikacijom i kvantifikacijom korištenjem tradicionalne mikroskopije kako bi se odredile
fitoplanktonske zajednice s dva različita lokaliteta u razdoblju od travnja do rujna 2019. godine.
Hipoteza ovog rada bila je da će u krškim jezerima u Hrvatskoj dominatne skupine fitoplanktona
biti Diatomeae, Chrysophyceae i Dinoflagellata. Sastav fotosintetskih pigmenata može poslužiti za
kemotaksonomsku procjenu fitoplanktonskih zajednica pa su iz tog razloga HPLC analizom
izmjerene različite koncentracije pigmenata (ß-karoten, lutein, fukoksantin) na temelju kojih su
određene dominantne skupine u ova dva prirodna krška jezera (Diatomeae, Chlorophyta i
Charophyceae). Usporedbom rezultata dobivenih metodom HPLC i standardnom mikroskopijom,
u oba jezera dokazana je prisutnost skupina Diatomeae i Chlorophyta kao dominantnih skupina,
dok se ostali rezultati nisu poklapali. Iako bi se u budućim istraživanjima trebali mjeriti još neki
karakteristični dijagnostički pigmenti kako bi se dokazala prisutnost ostalih skupina utvrđenih
mikroskopijom, u ovom istraživanju HPLC metoda se pokazala kao pouzdan i brz alat koji se, uz
mikroskopiju, može koristiti za brza istraživanja slatkovodnih ekosustava.

Abstract (english)

The presence of phytoplankton communities and distribution of photosynthetic pigments were
determined in two natural karst lakes, Lake Vrana near Biograd and Lake Vrana on Cres. The aim
of this work was to compare the results of high-performance liquid chromatography (HPLC
method) with taxonomic identification and quantification using traditional microscopy to
determine phytoplankton communities from two different localities in the period from April to
September 2019. The hypothesis was that in karst lakes in Croatia the dominant phytoplankton
groups will be Diatomeae, Chrysophyceae and Dinoflagellata. The composition of photosynthetic
pigments can be used for the chemotaxonomic assessment of phytoplankton communities, and for
this reason, different concentrations of pigments (ß-carotene, lutein, fucoxanthin) were measured
by HPLC. The dominant groups in these two natural karst lakes were Diatomeae, Chlorophyta and
Charophyceae. By comparing the results obtained by the HPLC method and standard microscopy,
the presence of Diatomeae and Chlorophyta groups as dominant groups was proven in both lakes,
while the other results did not match. Although in future research some more characteristic
diagnostic pigments should be measured to prove the presence of other groups determined by
microscopy, in this research the HPLC method proved to be a reliable and fast tool that, together
with microscopy, can be used for rapid research of freshwater ecosystems.
