Abstract

U okviru ovog diplomskog rada spektroskopski su okarakterizirane vodene otopine tri nova
derivata fenantridina s aminokiselinskim supstituentima (glicin, lizin) i
gvanidinokarbonilpirolnom jedinicom. Fenantridin je poznati fluorescentni marker za
polinukleotide i proteine dok aminokiselinski supstituenti pružaju dodatne mogućnosti za
interakcije s biološkim makromolekulama.
Konstante stabilnosti kompleksa ispitivanih spojeva s različitim DNA i RNA
polinukleotidima određene su metodom fluorescencijske spektroskopije. Način vezanja
peptidnih derivata fenantridina na DNA i RNA polinukleotide određen je eksperimentima
temperature mekšanja polinukleotida i spektroskopijom cirkularnog dikroizma. Konstante
stabilnosti kompleksa ispitivanih derivata s polinukleotidima izračunate su pomoću
Scatchardove jednadžbe metodom nelinearne regresije. Dobiveni rezultati ukazuju na vezanje
spojeva (najvjerojatnije u obliku dimera) u utore DNA i RNA te uzduž fosfatne okosnice
polinukleotida.

Abstract (english)

In the scope of this thesis, aqueous solutions of three new phenanthridine derivatives with
amino acid substituents (glycine, lysine) and a guanidinocarbonylpyrrole unit were
spectroscopically characterized. Phenanthridine is a known fluorescent marker for
polynucleotides and proteins. Amino acid substituents provide additional possibilities for
interactions with biological macromolecules. The stability constants of the complexes of the
studied compounds with different DNA and RNA polynucleotides were determined by
fluorescence spectroscopy. The binding mode of peptide phenanthridine derivatives to DNA
and RNA polynucleotides was determined by thermal melting experiments and circular
dichroism spectroscopy. The stability constants of the compound-polynucleotide complexes
were calculated by the Scatchard equation using the nonlinear regression method. The
obtained results indicate the binding of compounds (most likely in the form of dimers) in the
grooves of DNA and RNA and along the phosphate backbone of polynucleotides.
