Abstract

Stresna iskustva uključujući i ona doživljena tijekom osjetljivog razdoblja prenatalnog rasta povezuju se s disregulacijom HPA osi, koja rezultira s povišenim razinama kortizola u plazmi. Kohortna studija rođenih na istočnojadranskim otocima (CRIBS) proučava trudnice i njihovu djecu za utvrđivanje ranih odrednica zdravlja i bolesti, koje se javljaju još u fetalnom razdoblju i djetinjstvu. U ovom istraživanju proučeni su antropometrijski parametri majke i djeteta, razina kortizola u plazmi majki... More u drugom tromjesečju i metilacija 1F promotora NR3C1 gena na uzorku ovisno o geografskoj lokaciji (kopno-otok). Za analizu kortizola prilagođena je metoda tekućinske kromatografije visoke djelotvornosti. Metilacija je analizirana sangerovim sekvenciranjem nakon bisulfitne konverzije i umnažanjem specifičnog promotora. Kod ispitanica na otocima (Braču i Hvaru) u odnosu na kopno (Split i okolica), utvrđena je viša težina prije trudnoće, viša razina kortizola i veće razlike u porođajnoj težini, duljini i opsegu glave djece s obzirom na spol djeteta. Potvrđena je viša razina kortizola kod trudnica koje nose žensko dijete bez obzira na geografsku lokaciju. Ustanovljena je metilacija na 10 CpG mjesta promotorske 1F regije NR3C1 gena bez povezanosti s ostalim varijablama. Stressful experiences, including those experienced during the sensitive period of prenatal growth, are associated with dysregulation of the HPA axis, resulting in elevated plasma cortisol levels. The Eastern Adriatic Islands Birth Cohort Study (CRIBS) studies pregnant women and their children to explore early determinants of health and disease, which occur as early as the fetal period and childhood. More the mothers' plasma in the second trimester and the methylation of the 1F promoter of the NR3C1 gene were studied in the sample in relation to place of residence (mainland-island). A high-performance liquid chromatography method was adapted for cortisol analysis. Methylation was analyzed by Sanger sequencing after bisulfite conversion and amplification of the specific promoter. Among the subjects on the islands (Brač and Hvar) compared to the mainland (Split and its surroundings), a higher pre-pregnancy weight, a higher cortisol level and greater differences in the birth weight, length and head circumference of children with regard to the gender of the child were found. A higher cortisol level was confirmed in pregnant women carrying a female child regardless of their place of residence. Methylation was found at 10 CpG sites of the promoter 1F region of the NR3C1 gene without association with other variables.
