Abstract

Centrioli su valjkaste strukture okružene pericentriolarnom tvari koje izgrađuju centrosome, a centriol naslijeđen od majke ima ključnu ulogu u formiranju trepetljika. Potaknuti idejom Theodora Boveria o povezanosti centrosoma i raka, znanstvenici danas istražuju tu mogućnost pokušavajući otkriti što detaljnije osnovne mehanizme centrosoma, kao i povezanost s drugim bolestima. Bolesti koje se javljaju kao posljedica aberacija centriola često su rezultat nedostataka u samom nastanku centriola, u strukturi, funkciji i položaju centrosoma ili u formiranju i održavanju bičeva i trepetljika. Amplifikacija centrosoma je često svojstvo stanica raka jer potiče genetičku nestabilnost i nastanak tumora. Otkrivanje mutacija onkogena i tumorskih supresorskih gena može biti korisno u dijagnostici i praćenju liječenja. Svako otkriće na ovom području daje odgovore, nerijetko i otvara nova pitanja, ali nas u konačnici približava uspjehu u liječenju bolesti poput malignih tumora, kao i njihovom ranom otkrivanju.

Abstract (english)

Centrioles are barrel-shaped structures in matrix of protein (pericentriolar material), which form a core of centrosomes. They are important for forming cilia and flagella, as well. Following Theodor Boveri's idea of link between centrioles and human cancer, scientists are exploring that possibility and trying to find out details of basic mechanisms in centrosome and link with other diseases. Human diseases resulting from centriole aberrations reflect defects in centriole biogenesis, centrosome structure, function and positioning, or the formation and maintenance of cilia and flagella. Centrosome amplification is common feature of cancer cells driving genetic instability and tumorigenesis. Revealing oncogen mutations and tumor supressor genes can be useful in dignostic and treatment. Every finding in this area answers the questions, often asking new ones, but it is always a step forward to cure for tumors and their early discovery.
