Abstract

Provedena je opsežna metabolomička analiza uzoraka urina ispitanika oboljelih od raka testisa
i kontrolnih ispitanika. Hlapljivi organski metaboliti analizirani su vezanim sustavom plinske
kromatografije i spektrometrije masa uz Headspace mikroekstrakciju na čvrstoj fazi.
Dobiveni spektri masa identificirani su pomoću identifikacijskog sustava AMDIS (engl.
Automated Mass Spectral Deconvolution and Identification System), indeksa zadržavanja i
temperature vrelišta. Sustav AMDIS se pokazao kao pouzdan i moćan automatski sustav koji
ubrzava proces identifikacije. Među 128 na taj način identificiranih metabolita izabrano je 45
metabolita koji su zbog zadovoljavajuće stabilnosti i uloge u metabolizmu čovjeka uzeti u
obzir pri daljnjem traženju potencijalnih bioloških biljega tumora testisa.

Abstract (english)

A comprehensive metabolomic analysis of urine samples was performed in testicular cancer
patients and control subjects. Volatile organic metabolites (VOMs) were analysed by the
headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography – mass
spectrometry (GC-MS). Identification system AMDIS (Automated Mass Spectral
Deconvolution and Identification System), retention index and boiling temperature of
unknown compound were used for identification of unknown VOMs. Our results confirmed
the applicability of AMDIS for automated data evaluation as a reliable and powerful tool that
reduces the evaluation time. Among the 128 identified metabolites, 45 metabolites were
selected due to satisfactory stability and predicted role in human metabolism to be considered
in further investigation of potential biological tumour marker.
