Abstract

Cilj rada bio je odrediti utjecaj valporata na antioksidacijsku obranu u mozgu miša. Dodatni cilj bio je istražiti da li primjena polifenola naringenina zajedno sa valproatom utječe na redoks ravnotežu te može li sinergistička aplikacija naringenina i valproata umanjiti potencijalne negativne učinke. Pokusima je izmjerena lipidna peroksidacija, koncentracija ukupnog glutationa te aktivnost antioksidativnog enzima superoksid dismutaze u prefrontalnom i centralnom korteksu i malom mozgu. Rezultati su pokazali da, tretman valproatom, naringeninom i njihovim kombinacijama uzrokuje povećanu lipidnu peroksidaciju i kod mužjaka i kod ženki u sve tri mjerene regije mozga. Protektivni učinak polifenolne molekule naringenina nije pokazan. Sukladno povećanoj lipidnoj peroksidaciji raste i aktivnost superokisd dismutaze u svim regijama mozga. Izrazito snižena koncentracija reduciranog glutationa u mozgu nastala je vjerojatno zbog povećanog superoksidnog radikala i oksidacijske ravnoteže u stanici te je glutation prešao iz reduciranog u oksidirani oblik.

Abstract (english)

Aim of this study was to determine effects of antioxidative defense in mouse brain (C57Bl6). An additional aim was to investigate whether the use of polyphenylene naringenine together with valproate affects the redox balance and whether the synergistic application of naringenine and valproate can reduce the potential negative effects. In this experiment lipid peroxidation, concentration of total glutathione and antioxidant activity of superoxide dismutase was measured in the prefrontal and central cortex and small brain. Results showed that the treatment with valproate and naringenine and their combinations caused increased lipid peroxidation in both males and females in all three measured brain regions. The protective effect of polyphenolic naringenine molecule has not been demonstrated. In accordance with increased lipid peroxidation, activity of superoxide dismutase in the brain has also increased. Reduced concentration of reduced glutathione in the brain was probably due to the increased superoxide radical and oxidation equilibrium in the cell and glutathione ranged from reduced to his oxidized form.
