Abstract

Elvitegravir je inhibitor HIV-1 integraze koji se u kombinaciji s drugim lijekovima koristi za liječenje infekcije virusom humane imunodeficijencije (HIV). Inhibiranjem enzima integraze, elvitegravir smanjuje količinu HIV-a u tijelu, te spriječava njegovo daljnje razmnožavanje. Ispitivanje čistoće lijeka uključuje analitičke metode kojima je cilj detekcija, identifikacija, strukturna karakterizacija i kvantitativno određivanje onečišćenja. U ovom diplomskom radu razvijena je brza i učinkovita metoda tekućinske kromatografije ultravisoke djelotvornosti (UHPLC) za analizu acetamida u elvitegraviru. S obzirom da je acetamid molekula male molekulske mase, koristile su se tehnike tekućinske kromatografije ultravisoke djelotvornosti uz detekciju detektorom s nizom dioda (UHPLC-DAD) i trostrukim kvadrupolom (UHPLC-QQQ). Metode razvijene u diplomskom radu su validirane.

Abstract (english)

Elvitegravir is an HIV-1 integrase inhibitor, which is used in combination with other active pharmaceutical ingredients as a treatment of the human immunodeficiency virus (HIV). By inhibiting enzyme integrase, elvitegravir is lowering the amount of HIV in the human body and inhibits its further propagation. Analysis of pharmaceutical impurities includes analytical procedures with aim to detect, identify, structurally characterize and quantify the impurity. In present study we developed fast and reliable method of ultra-high performance liquid chromatography for determination of acetamide in elvitegravir. Acetamide is a molecule of low molar mass, that is why we used the diode array detector (UHPLC-DAD) and triple quadrupole detector (UHPLC-QQQ). Methods developed in this research work are validated.
