Oštećenja molekule DNA, koja nastaju kao posljedica izlaganja toksičnim tvarima, mogu u konačnici utjecati na zdravlje i preživljavanje organizama. Stoga je bitno razvijati i testirati osjetljive molekularno citogenetičke metode za ranu detekciju i procjenu takvih promjena. Cilj ovog rada bio je procijeniti oštećenja molekule DNA u zebricama (Danio rerio) izloženim modelnim genotoksikantima (benzo[a]piren (B[a]P), etilmetanosulfonat (EMS)) te utvrditi dugotrajnost takvih oštećenja. Primarno... More oštećenje DNA detektirano je metodom Komet-testa dok su dugotrajne posljedice praćene metodom umnoženih polimorfnih DNA biljega (engl. amplified fragment length polymorphism, AFLP). Rezultatima ovog istraživanja dokazana su primarna oštećenja DNA uzrokovana modelnim genotoksikantima te perzistentnost promjena u genomu zebrica i godinu dana nakon izlaganja genotoksikantima, te je potvrđena pogodnost korištenja zebrica kao modelnih organizama u istraživanjima genotoksičnosti. Abstract (english)

DNA damage as a consequence of toxicant exposure may in the long run influence health and survival of the organisms. Therefore, it is of paramount importance to develop and test sensitive molecular cytogenetic methods for early detection and assessment of these changes. The aim of this study was to assess DNA damage in zebrafish (Danio rerio) exposed to model genotoxicants (Benzo[a]pyrene (B[a]P), Ethyl methanesulfonate (EMS)) and to determine the longevity of such damage. The primary DNA... More damage was detected using Comet assay whereas the long-term effects were monitored using the AFLP (amplified fragment length polymorphism) method. The results of this study demonstrated the primary DNA damage caused by model genotoxicants as well as persistence of alterations in the zebrafish genome even one year after the end of the genotoxic exposure.
