Abstract

U okviru ovog diplomskog rada provedena je spektroskopska karakterizacija otopina spojeva CT-A1, CT-A2, CT-A4 i CT-A14, ispitana je njihova interakcija s dvolančanim polinukleotidima i potencijalno antioksidacijsko djelovanje. UV/Vis spektri spojeva snimljeni su u području od 230 do 600 nm i odreĎene su vrijednosti molarnih apsorpcijskih koeficijenata () pri valnoj duljini maksimuma apsorpcije (max). UtvrĎeno je da su izmjerene apsorbancije puferskih otopina istaţivanih spojeva proporcionalne njihovim koncentracijama što je u skladu s Lambert – Beer-ovim zakonom. Ispitana je ovisnost intenziteta fluorescencije o koncentraciji spojeva u području od 450 do 650 nm te su u tom koncentracijskom rasponu provedene fluorimetrijske titracije s dvolančanim polinukleotidima. U fluorimetrijskim titracijama s dvolančanim polinukleotidima došlo je do povećanja fluorescencije spojeva CT-A1 i CT-A2 za razliku od smanjenja fluorescencije spoja CT-A14 dok su kod spoja CT-A4 promjene intenziteta emisije u većini titracija bile neznatne. Općenito, spojevi su pokazali veći afinitet prema dvolančanim polinukleotidima DNA nego RNA. Rezultati CD titracija DNA polinukleotida s gotovo svim ispitivanim spojevima pokazali su slabe negativne CD (ICD) signale na valnoj duljini oko 300 nm. Ovi rezultati upućuju na interkalativan način vezanja. Metodoma ABTS i DPPH utvrĎena je značajna antioksidacijska aktivnost svih ispitivanih spojeva osim spoja CT-A14.

Abstract (english)

In this thesis, spectroscopic characterization of solutions of CT-A1, CT-A2, CT-A4 and CT-A14 compounds was performed, their interaction with double-stranded polynucleotides and potential antioxidant activity were examined. The UV/Vis spectra of the compounds were recorded in the range from 230 to 600 nm and (max) the values of molar absorption coefficients () were determined at the wavelength of maximum absorption. The absorbance of buffered solutions of the newly synthesized compounds was found to be propotional to their concentrations, which is in accordance with the Lambert – Beer law. The dependence of fluorescence intensity on the concentration of the compounds was tested in the range of 450 to 650 nm and in this concentration range fluorimetric titrations with double-stranded polynucleotides were performed. The results of fluorimetric titrations with double-stranded polynucleotides showed an increase of fluorescence of CT-A1 and CT-A2 compounds as opposed to a decrease of fluorescence of CT-A14 compound, whereas fluorescence changes of CT-A4 compound in most titrations were negligible. In general, the examined compounds showed higher affinity toward ds-DNA polynucleotides than ds-RNA. Results of CD titrations of DNA polynucleotides with almost all tested compounds showed weak negative CD (ICD) signals located around 300 nm. These results suggest an intercalative way of binding. The significant antioxidant activity measured by ABTS and DPPH methods was found for all tested compounds except for CT-A14.
