Abstract

Kristalizacijom inzulina s metalnim ionima (bakar(II), nikal(II) i kobalt(II)) u prisustvu različitih koncentracija halogenidnih iona (Cl-, Br- i I-)dobiveno je 28 različitih jediničnih kristala koji su analizirani rendgenskom difrakcijom. Riješene su kristalne strukture triju derivata inzulina. Niklov(II) derivat inzulina kristaliziran u prisustvu bromidnih iona (c = 0,898 mol dm-3) kristalizira u prostornoj grupi H32 s jednim monomerom inzulina u asimetričnoj jedinici dok u prisustvu niže koncentracije bromidnih iona (c = 0,148 mol dm-3) kristalizira u prostornoj grupi H3 s jednim dimerom inzulina u asimetričnoj jedinici. Kobaltov(II) derivat inzulina kristaliziran u prisustvu kloridnih iona (c = 0,248 mol dm-3) također kristalizira u prostornoj grupi H3, ali s dva dimera inzulina u asimetričnoj jedinici. U svim analiziranim strukturama heksameri inzulina su T6 forme te sadrže dva metalna iona (Ni/Co) koordinirana s tri dušikova atoma iz simetrijski ovisnih molekula histidina i tri kisikova atoma iz tri molekule vode, ali su različite stabilnosti.

Abstract (english)

Crystallization of insulin with metal ions (copper(II), nickel(II) and cobalt(II)) in the presence of different concentrations of halide ions (Cl-, Br- i I-) yielded 28 different single crystals which were analyzed by X-ray diffraction method. The crystal structures of three insulin derivatives were solved. Nikel(II) insulin derivative crystallized in the presence of bromide ions (c = 0,898 mol dm-3) crystallizes in the H32 space group with one insulin monomer in the asymmetric unit, while in the presence of lower concentration of bromide ions (c = 0,148 mol dm-3) crystallizes in the H3 space group with one dimer in the asymmetric unit. The cobalt(II) insulin derivative crystallized in the presence of chloride ions (c = 0,248 mol dm-3) also crystallizes in the H3 space group, but with two dimers in the asymmetric unit. In all analyzed structures, insulin hexamers are in T6 form and two metal ions (Ni/Co) are octahedrally coordinated by three symmetry-related nitrogen atoms of three histidine molecules and three oxygen atoms from three water molecules, but with different stability.
