Abstract

Stanično okruženje i međustanična komunikacija su jedan od bitnih čimbenika u odgovoru stanica na stres. U 3D kulturama stanice su u okruženju koje je najsličnije uvjetima u tkivima, dok u 2D kulturama stanice rastu na podlozi u jednom sloju i ne stvara se izvanstanični matriks. Jedan od tipova međustanične komunikacije su premošćujuće strukture koje nastaju između stanica. To su tunelirajuće nanocjevčice (engl. tunneling nanotubes, TNT) i tumorske mikrocjevčice (engl. tumor microtubes, TM). U ovom radu opisane su premošćujuće strukture u stanicama U2OS. Utvrđeno je da tretman hladnoćom povećava broj premošćujućih struktura u uzorcima dok tretman inhibitorom proteina sHSP27 J2 utječe na smanjenje broja premošćujućih struktura. Također, zabilježena je lokalizacija proteina sHSP27 i membranskih struktura koja upućuje na njihov transport. Uspostavljen je protokol za izradu 3D kultura metodom magnetske levitacije stanica. Uspoređen je izgled stanica u 2D i 3D staničnom okruženju i razina oksidativnog stresa mjerena bojom za detekciju stresa CellROXTM Deep Red Reagent.

Abstract (english)

The cell environment and intercellular connections have major impact in cellular stress response. Cell environment in 3D cultures is the most similar to the one in tisssues, while at 2D cultures cells grow on substrate in a single layer and don’t produce extracellular matrix. One type of cell to cell connections are structures that interconnect the cells. These are tunneling nanotubes (TNTs) and tumor microtubes, (TMs). In this paper, tunneling structures in U2OS cells are described. Cold shock treatment was found to increase the number of TNTs and TMs, while the treatment with sHSP27 inhibitor J2 reduced the number of structures. Also, localization of protein sHSP27 and membrane structures that refer to the transport is noted. A protocol for making 3D cultures by magnetic levitation has been established. The appearance of cells in a 2D and 3D cell environment and the level of oxidative stress measured by CellROXTM Deep Red Reagent stress detection were compared.
