Abstract

U okviru ove doktorske disertacije provedena je asocijacijska studija funkcionalno najvažnijih polimorfizama četiri kandidatska «gena dugovječnosti» (TP53, IL-6, TNF-α i SIRT1) sa širokim spektrom varijabli (anketnih i biometrijskih) povezanih sa zdravljem u uzorku od 344 osobe duboke starosti (85 i više godina). Univarijatne analize istaknule su spolno specifičnu strukturu povezanosti varijabli zdravstvenog statusa s ispitivanim polimorfizmima. Kod oba spola, faktorska analiza izlučila je faktor „općeg zdravlja“ koji je uz a priori konstruirane aditivne skale zdravstvenih rizika korišten kao varijabla biološke dobi. Među pet ispitivanih polimorfizama samo je rs1800795 polimorfizam IL-6 gena pokazao značajnu povezanost s kompozitnim varijablama biološke dobi kod muškaraca te time svoj mogući pleiotropni utjecaj na proces starenja kod ovog spola. S obzirom na dostupnost literaturnih podataka iz različitih populacija, međugeneracijska meta-analiza provedena je za dva polimorfizma (u TP53 i IL-6) te je indicirala mogući utjecaj Arg72Pro polimorfizma TP53 gena na dugovječnost.

Abstract (english)

In this dissertation an association study of functionally most important polymorphisms in four candidate longevity genes (TP53, IL-6, TNF-α, SIRT1) was conducted with a wide spectrum of variables associated with health status in 344 people of very old age (85+ yrs). Univariate analysis highlighted gender specific association of health status variables with investigated polymorphisms. In both genders factor analysis extracted the "general health" factor which was used as a biological age variable together with a priori constructed additive health risks scale. Among five investigated polymorphisms only the rs1800795 polymorphysm of IL-6 gene showed significant association with composite variables of biological age in male population, and thereby its pleitropic effect on the process of ageing in male sex. Considering availability of literature data for different populations, intergenerational meta-analysis was conducted for two polymorphisms (in TP53, IL-6) and indicated the possible effect of Arg72Pro polymorphism of TP53 gene on longevity.
