Povezanost polimorfizama gena za leptin i leptinski receptor s pretilošću u ljudi

Abstract

Pretilost je složeni poremećaj koji nastaje zbog neravnoteže unosa i potrošnje energije, te je bitan rizični faktor za velik broj kroničnih bolesti. Signalni put leptina jedan je od najbitnijih puteva uključenih u regulaciju energetske homeostaze. Leptin je hormon koji se primarno izlučuje u bijelom masnom tkivu. Leptinski receptori se nalaze u mnogim organskim sustavima, ali posebno su važni oni u hipotalamusu gdje reguliraju energetsku homeostazu (unos hrane i potrošnju energije). Opisani su brojni polimorfizmi u genima za leptin i leptinski receptor kod ljudi, te se za neke smatra da su povezani s razvojem pretilosti. U ovom
istraživanju se na 375 uzoraka DNA Hrvatskih Roma ispitala povezanost polimorfizama Gln223Arg i Lys109Arg leptinskog receptora i 27 polimorfizama dijela promotorske regije leptinskog gena s pretilošću. Samo je polimorfizam Lys109Arg (genotip AA) pokazao povezanost s prekomjernom težinom kod muškaraca, dok polimorfizmi promotorske regije leptina i polimorfizam Gln223Arg nisu pokazali nikakvu povezanost s pretilošću.

Abstract (english)

Obesity is complex disorder which develops when energy intake exceeds energy utilization. It increases the risk for a variety of chronic diseases. The leptin-signalling pathway is considered one of the most important routes involved in energy homeostasis. Leptin is a hormone produced predominantly by the white adipose tissue. Leptin receptor is expressed in many tissues, but it is particularly important in the hypothalamus, where it regulates energy homeostasis (food intake and energy expenditure). Many leptin and leptin receptor genes polymorphisms have been investigated, and some of them are candidates for association with obesity. In this study, leptin receptor polymorphisms Gln223Arg and Lys109Arg, and 27 leptin promoter polymorphisms have been analysed for association with obesity, in 375 Croatian Roma DNA samples. The results indicate that there is no association between Gln223Arg, and leptin promoter polymorphisms with obesity. However, Lys109Arg AA genotype is associated with overweight in men.
