Abstract

Oscilacije kinetohora primijećene su u različitim organizmima od kvasca do sisavaca. Te se oscilacije javljaju dok stanica provjerava jesu li sve kinetohore ispravno spojene, što je poznato kao kontrolna točka diobenog vretena. Stoga je važno razumjeti mehanizam oscilacija kinetohora kako bi se razjasnilo kako stanica regulira sile napetosti između sestrinskih kinetohora. Oscilacije kinetohora ovise o dinamici mikrotubula koja uključuje rast mikrotubula i njihovo skraćivanje, a time i o specifičnim faktorima koji upravljaju vjerojatnošću da se mikrotubul nađe u stanju rasta ili skraćivanja. Cilj ovog istraživanja je pokazati na koji način se mijenja gustoća motora na mikrotubulima korištenjem Laplaceovog transformata te derivacijom pomoću metode konačnih razlika. Nakon toga se izrađuje model oscilacija kinetohora - jedan fizikalni i jedan maksimalno pojednostavljeni, te se uspoređuje ponašanje ta dva modela za različite sustave mikrotubula i kinetohora.

Abstract (english)

Kinetochore oscillations have been observed in a variety of organisms, from yeast to mammals. These oscillations occur while the cell checks whether all the kinetochores are properly merged, which is known as the mitotic checkpoint. Therefore, it is important to understand the mechanism of kinetochore oscillation to clarify how cells regulate the tension force between sister kinetochores. Kinetochore oscillations depend on the microtubule dynamics which involves the growth and shrinkage of the microtubule, and thus on the specific factors that control the probability that the microtubule finds itself in a state of growth or shrinkage. The aim of this study is to show how the motor density is changing by using the Laplace transform and by derivation using finite difference methods. Afterwards, two kinetochore oscillation models are created. One physical and one completely simplified. The models are then compared for different microtubule and kinetochore systems.
