Abstract

Karcinom debelog crijeva treći je zloćudni tumor po učestalosti u svijetu i na drugom je mjestu uzroka smrtnosti od raka. Nastaje kao posljedica stupnjevitog procesa tijekom kojeg iz dobroćudnog adenoma nakupljanjem mutacija nastaje zloćudni invazivni tumor. Postojeći prognostički parametri još uvijek samo djelomično daju informaciju o toku i ishodu ove bolesti, stoga postoji potreba za pronalaskom novih dijagnostičkih i prognostičkih molekularnih biljega. Jedan od tih biljega mogao bi biti i citokin IL-1β. Povišene vrijednosti citokina dokazane su u različitim tipovima tumora. Dosadašnja istraživanja pokazala su povezanost polimorfizama -31 T/C te +3954 C/T gena IL-1β s pojačanom ekspresijom i povišenim serumskim vrijednostima IL-1β u čovjeka. Cilj diplomskog rada bio je ispitati povezanost prethodno navedenih polimorfizama gena IL-1β sa sklonošću obolijevanju i preživljenjem oboljelih od sporadičnog karcinoma debelog crijeva. Rezultati istraživanja pokazali su da postoji povezanost polimorfizma +3954 C/T gena IL-1β s preživljenjem oboljelih od sporadičnog karcinoma debelog crijeva.

Abstract (english)

Colon cancer is the third most common cancer worldwide and the second leading cause of cancer death. It occurs as a consequence of multistep process in which colorectal carcinoma develops from adenoma, as a result of mutations accumulation. Existing prognostic markers give only partial information about course and outcome of this disease, so there is a need for discovery of novel disease-related diagnostic and prognostic molecular markers. One of these new markers could be also cytokine IL-1β. A number of studies have reported increased IL-1β concentration in different cancer types. Previous studies have shown association between -31 T/C and +3954 C/T IL-1β gene polymorphisms with higher expression and elevated IL-1β serum levels. The aim of this study was to investigate the correlation between these two IL-1β gene polymorphisms and susceptibility to disease and survival of patients suffering from sporadic colorectal cancer. Our results showed the correlation between polymorphisms +3954 C/T and survival of patients with colon cancer.
