Abstract

U radu su prvi put istraženi detalji elektrokemijske analize modelnog aminopolisaharida
kitozana, proteina goveđeg serum albumina (BSA), te njihove smjese u morskoj vodi.
Mjerenja su izvedena na visećoj živinoj kapi koristeći adsorptivnu kronopotenciometriju s
pobudnom strujom (CPS). Oba analita dala su karakterističan pik H (Ep = ~ –1.8 V) kao
posljedicu katalitičke redukcije vodikovih iona. Iako je samo BSA bilo moguće detektirati in
situ u morskoj vodi, za oba analita je najosjetljivija detekcija bila nakon adsorptivnog
prijenosa (AdT) iz morske vode u fosfatni pufer pH ~ 7,0. Stoga su definirani optimalni
eksperimentalni uvjeti za njihovu AdT CPS analizu: potencijal akumulacije, Ea= –0,1 V,
vrijeme akumulacije, ta = 60 s, te pobudna struja, Istr = –20 μA za kitozan te Istr = –40 μA za
BSA. AdT CPS detekcijom smjese dobiven je samo jedan pik H čime selektivna detekcija
kitozana, a ni BSA u njihovoj smjesi nije bila moguća.

Abstract (english)

The thesis explored for the first time the details of electrochemical analysis of chitosan
aminopolysaccharide, BSA protein, and their seawater mixture. Measurements were
performed on a hanging mercury drop electrode by using constant current
chronopotentiometric stripping (CPS). Both analytes gave a characteristic peak H (Ep = ~ –1.8
V) due to catalytic hydrogen evolution reaction. Although only BSA can be detected in situ in
seawater, the most sensitive detection for both analytes was after an adsorptive transfer (AdT)
from seawater to phosphate buffer pH ~ 7.0. Therefore, optimal experimental conditions for
their AdT CPS analysis were defined: accumulation potential, Ea = –0.1 V, accumulation
time, ta = 60 s, and stripping current, Istr = –20 μA for chitosan and Istr = –40 μA for BSA.
Only one AdT CPS peak H was observed in mixture, so selective detection of neither chitosan
nor BSA in their mixture was possible.
